摘要
目的体外研究化学药物对胃癌细胞株 MKN-45的细胞周期影响。方法应用Brdu/DNA 双标记 FCM 技术,观察多种化疗药物对 MKN-45细胞周期动力学的影响。结果 5-Fu、VP16和 MMC 能显著影响肿瘤细胞的周期动力学,阻抑肿瘤细胞生长;甲酰四氢叶酸能加强5-Fu 的作用。而加用阿霉素、顺氨氯铂并不加强5-Fu 或 MMC 的作用。结论研究了化疗药物对胃癌细胞株的细胞周期动力学改变,为临床化疗药物应用提供参考数据。
Objective We investigate the effects of chemotherapeutic agents on the cell cycle of gastric carcinoma,MKN-45 cell line,in vitro.Methods The effects of several chemotherapeutic agents on cell cycle kinetics of MKN-45 were observed by means of Brdu/DNA double-labelling technique.Results 5-Fu,VP16, and MMC influenced significantly the cell cycle kinetics of tumor cell,and inhibit the cell growth;leucovorin increased the effect of 5-Fu,whereas doxorubicin, cisplatin did not increase the effect of 5-Fu or MMC.Conclusion The present study provides references data for clinical use of chemotherapeutic agents.
