摘要
采用高效液相色谱法测定不同生长年限杜仲皮中绿原酸的含量。色谱柱:KromasilC18(250mm×4.6mm,5μm),流动相:乙腈-0.4%磷酸(13:87),检测波长:327nm,流速1mL/min,柱温:25℃。结果表明:杜仲内皮中绿原酸含量以22年生最高,栓皮中绿原酸含量以13年生最高。从杜仲皮绿原酸总含量考虑,13年左右为最佳采收期。本实验建立的杜仲皮绿原酸含量测定方法,重现性好,结果准确可靠,为杜仲皮质量控制提供了参考。通过对不同生长年限杜仲皮中绿原酸含量的测定,为杜仲的合理采收期提供了科学依据。
This article aimed to determinate the content of chlorogenic acid in the bark of Eucommia of different ages by HPLC. The method adopted a Kromasil C,s column(250 mm ×4.6 mm,5 μm) ,with mobile phase consisted of acetonitrile-0.4% Phosphoric acid ( 13: 87), at a flow rate of 1.0 mL/min; determination at 327 nm, and 25 ℃. The results indicated that the content of chlorogenic acid in 22 ages inside bark of Eucommia is the highest. The content of chlorogenic acid in 13 ages outside bark of Eucommia is the highest. If only considering the total content of chlorgenic acid,the best harvest time of Eucommia is 13 years. This method was accurate and credible. The results provided the scientific basis for the harvest of Eucommia.
