摘要
目的探讨p53基因和端粒酶活性在良、恶性梗阻性黄疸中的表达。方法选取恶性梗阻性黄疸患者64例作为实验组,良性梗阻性黄疸患者60例作为对照组,采用端粒重复序列扩增-聚合酶链式反应-酶联免疫吸附测定法检测端粒酶活性,流式细胞仪检测p53蛋白的阳性表达率。结果恶性和良性梗阻性黄疸组织标本中端粒酶表达阳性率分别为87.50%和3.33%,p53蛋白阳性表达率分别为71.88%和0。结论端粒酶活性和p53蛋白阳性表达与淋巴转移率和肿瘤的分化程度有明显关系。端粒酶和p53基因可以作为胆、胰系统恶性肿瘤诊断的有力标志物,联合检测可以提高恶性梗阻性黄疸的确诊率。
Objective To investigate the expression of telomerase and p53 gene in biliary-pancreatic system cancer. Methods 64 patients with malignant biliary Obstruction were selected as experimental group and 60 patients with benign biliary obstruction as control group. The telomerase activity was tested with telomere repeat amplification protocol-polymerase chain reaction-enzyme linked immunosorbent assay and the expression of p53 was tested by flow cytometry. Results The telomerase activity and p53 positive rate of patients with malignant obstructive jaundice were 87.5% and 71.88%. Correspondingly, they were 3.33% and 0 in benign biliary obstruction. Conclusion Testing the expressions of telomerase and p53 can improve the accuracy and sensitivity in diagnoses of malignant biliary obstruction.
