摘要
目的:探讨云芝多糖(CVP)对免疫性肝损伤的保护作用及其机制。方法:昆明种小鼠随机分为5组,分正常对照组;病理造模组:CVP保护组分高、中、低三组。造模第一天,除正常对照组外,其余各组小鼠经尾静脉注射2.5 mg卡介苗(BCG);CVP低、中、高保护组每天分别按体重100 mg/kg、200m g/kg、300 mg/kg CVP灌胃1次,正常对照组和模型组用生理盐水同样处理,第十二天用CVP灌胃给药2 h后,模型组和CVP各保护组从尾静脉注射7.5μg/只脂多糖(LPS);16 h后称取小鼠体重,眼球取血分离血清测定丙氨酸转氨酶(ALT);处死小鼠立即取出肝脏、胸腺称重,计算肝体指数和胸腺指数,制备肝匀浆测定丙二醛(MDA),取相同部位的肝组织,制作病理切片,HE染色,光镜下观察肝组织损伤程度。结果:与模型组比较,CVP各保护组均可降低免疫性肝损伤小鼠肝体指数,增加胸腺指数;降低小鼠血清中ALT的活性和和肝匀浆中MDA的含量,减轻肝细胞气球样变性,点状、片状坏死及炎性细胞浸润。结论:CVP对小鼠免疫性肝损伤有保护作用。
Objective :To investigate the Protective effect of coriolus versiclor polysaccharides (CVP) on immunological liver injury and its meehanism.Methods:Kunming mice were randomly divided into 5 groups, sub-normal control group;pathological model;PSK protection component high, medium and low three groups.on the first day, in addition to the normal control group, the remaining mice in each group received via the tail vein injection of 2.5 mg BCG;CVP low,middle and high protection groups took oral CVD in a dose of 100 mg/ kg,200 mg/kg,300 mg/kg respectively once daily.The normal control group and model group received,the same treatment with saline.On 12d at 2 h after oral administration,each mouse in the model group and the protection groups was given from the tail vein injection of 7.5 bμg LPS; 16 h later the mice were weighed,blood was ealleeted from eyeball for detecting serum ALT;the mice were killed immediately for removing liver, thymus to weigh to calculate the liver somatic index and thymus index.Then to prepare liver homogenate for deter. mining MDA.From the same parts of the liver tissue,to make pathologic sections and HE staining for observing liver tissue damage by lighe microscope.Results: Compared with model group, CVP could reduce mice liver somatieindex in protective immunological liver injury,increase thymie index,lower serum ALT activity in liver homogenate and MDA content of liver cells and reduce the balloon degeneration, punctate, patchy necrosis and inflammatory cell infiltration.Conclusion :CVP has a protective effect on immunological liver injury in mice.
