Expression and clinical significance of erythropoietin and erythropoietin receptor in tongue squamous cell carcinoma

摘要 目的探讨红细胞生成素(Epo)和红细胞生成素受体(EpoR)在舌鳞状细胞癌(TSCC)中的表达水平及其与微血管密度的关系,并结合临床病理资料评价其临床意义。方法采用免疫组织化学法检测具有完整临床病理资料的65例TSCC组织中Epo和EpoR表达情况,并同时检测其微血管密度,以50例癌旁组织作为对照。分析Epo和EpoR表达水平对TSCC患者预后的评估价值。结果TSCC组织中Epo和EpoR的表达高于癌旁组织(P<0.05)。Epo的表达与患者的年龄、微血管的密度、肿瘤分期相关(P<0.05)。EpoR的表达与微血管的密度相关(P<0.05)。Kaplan-Meier生存分析显示Epo和EpoR阳性表达组患者总体生存率显著低于其阴性表达组。多因素Cox回归分析表明Epo和EpoR表达水平是TSCC患者预后判断的重要因子(P<0.05)。结论Epo和EpoR在TSCC发生发展及肿瘤微血管形成过程中起到重要作用,可能是评估TSCC患者预后的潜在标志物。 Objective To investigate the expression pattern and clinical pathological significance of erythropoietin/erythropoietin receptor (Epo/EpoR) in squamous cell carcinoma of the tongue (TSCC). Methods The level of Epo, EpoR and Microvessel Density (MVD) in 65 TSCC tissues was tested by immunohistochemistry. 50 para-cancer tissue were used as control. The level of Epo, EpoR expression and their effect on the prognosis was analyzed. Results The proportion of Epo and EpoR alterations in TSCC tissue was higher than that in normal squmous epitheliums around carcinoma (P 〈 0.05). Epo expression was associated with age, microvessel density, clinical staging (P 〈 0.05). The expression of EpoR was assciated with microvessel density (P 〈 0.05). The survival rate of patients with high Epo/ EpoR expression level is lower than that of those with low Epo/EpoR expression level. The results of Cox regression analysis revealed that levels of Epo and EpoR expressions are critical factors affecting postoperative survival rate (P 〈 0.05). Conclusions Epo and EpoR system provides advantage to the carcinogenesis, angiogenesis, and malignant progression of tongue sqamous cell carcinoma, and reconfirm Epo and EpoR as potential prognostic markers.
