摘要
目的:将超滤、纳滤技术应用于β-环状糊精生产中,得到无溶剂法生产β-环状糊精新工艺。方法以5%木薯淀粉和β-环状糊精葡萄糖基转移酶为原料,于85℃、pH 6.5进行淀粉的糊化和液化,在55℃、pH 7.0完成环化,生成β-环状糊精。选择不同截留分子量的膜组件,研究在不同膜组件分离过程中的分离条件,完成无溶剂法生产β-环状糊精。结果利用截留分子量为500Dalton的纳滤膜和截留分子量为10000Dalton的超滤膜联用,分离出酶促反应过程中生成的β-环状糊精,保证产品中没有溶剂溶剂的残留,真空干燥得成品,最终收率为33.63%。结论把膜分离技术应用于β-环状糊精生产,可以得到无溶剂法生产β-环状糊精的新工艺。
Aim Using UF and NF,we developed a new technology of solvent-free production of β-cyclodextrin.Methods cassava starch(5%) and β-cyclodextrin glucanotransferase were gelatinized and liquefied under the conditions of 85 ℃,pH6.5.The reaction of β-cyclodextrin was taken at 55 ℃,pH7.0.The different membranes was selected and its separation was studied.Result the ultrafiltration membrane with molecular mass cut-off is 10000 and the nanofiltration membrane with molecular mass cut-off 500 was selected.Through the UF and NF,β-cyclodextrin was completely extracted without any organic solvents.The yield was 33.63% by vacuum drying,Conclusion Use membrane separation technology in β-cyclodextrin production,β-cyclodextrin can be prepared without organic solvents.
