摘要
目的建立大鼠中空管线栓大脑中动脉(MCA)梗塞模型,并观察经中空管灌洗冷生理盐水达到局部低温的效果。方法30只大鼠,分为经典线栓组、中空管线栓组和低温盐水灌洗组。后者是在血液再灌注前10 min,通过特制的中空管注入6 mL 20℃生理盐水到MCA缺血区。结果48 h后中空管线栓组脑梗死体积(41.81±2.88)%与经典线栓组(41.92±2.17)%相比差异无显著性意义(P=0.922)。经中空管注入冷生理盐水,供血区皮质温度从(37.0±0.1)℃降低至(32.8±0.2)℃,髓质温度从(37.5±0.1)℃降低至(33.2±0.3)℃;脑梗死体积(22.37±1.86)%同中空管线栓组相比,差异有统计学意义(P<0.01)。结论用特制中空管替代传统线栓,能够制作稳定的MCA梗塞模型。在血液再灌注之前,经动脉内注入冷生理盐水,能够诱导缺血脑组织局部轻度低温,并减少脑梗死体积。
Objective To establish a new rat model of middle cerebral artery(MCA)ischemia with modified hollow thread and to observe the effectiveness of intra-arterial cold saline injection to induce local hypothermia. Methods Thirty rats were divided into three groups. MCA ischemia with solid thread was induced in the rats of the first groups. MCA ischemia with modified hollow thread was induced in the rats of the second group. In the rats of the last group,6 mL of 20℃ normal saline solution was flushed at a speed of 0.6 mL per rain, beginning 10 min before the blood reperfusion. The infarct volume of rat brains stained by TTC was observed 48 h later. The brain temperature, the blood flow and the neurological scores were recorded during this procedure. Results The infarction volume in the second group with modified hollow thread[(41.81±2.88)%]was similar to that in the second group with solid threadE(41.92±2.17)%,P=0. 922]. The cold saline(20℃)infusion via the MCA rapidly reduced the temperature of the MCA supplied ischemic territory in cortex from (37.0±0.1)℃ to (32.8±0.2)℃ and in striatum from (37.5±0. 1)℃ to (33.2±0.3)℃. The infarction volume of the third group was (22.37±1.86) %0 ,which was significantly less than that in the second group without cold saline infusion(P〈0.01). Conclusion A new rat MCA ischemia model can be established with modified hollow intra arterial thread. Local brain hypothermia can be reduced with cold saline infusion before blood reperfusion and can decrease the infarction volume.
