摘要
目的探讨不同气腹介质对结直肠癌肿瘤转移的影响。方法体外培养人结直肠癌HT-29细胞株,模拟CO2气腹、He气腹处理后,通过MTT法检测细胞的黏附性;应用免疫组化检测E-钙黏素的表达。建立气腹动物实验模型,观测腹腔转移瘤的情况。结果模拟CO2气腹组细胞黏附力大于模拟He气腹组、对照组;模拟CO2气腹组E-钙黏素的表达小于模拟He气腹组、对照组;模拟He气腹组与对照组两者差别无统计学意义。CO2气腹组裸鼠移植瘤平均瘤重大于He气腹组、免气腹组;He气腹组与免气腹组差别无统计学意义。结论CO2气腹有可能对肿瘤转移起一定促进作用,He气腹对肿瘤转移无明显影响。
Objective To evaluate the effects of different artificial pnettmoperitoneum medium on colorectal cell metastasis, to explore the safety of colorectal carcinoma metastasis in laparoscopic colorectal carcinoma operation. Methods With an in vitro artificial pneumoperitoneum model, HT-29 human colon carcinoma cells were exposed to CO2-insuffllation, He-insuffiation and control group. The adhesion of carcinoma cells were detected by MTT; the expression of E-cadherin in carcinoma cells were examined by the immunohistochemical method. In the in vivo study, the weight of metastases were measured in each mouse. Results The adhesion of simulated CO2 pneumoperitoneum group was stronger than simulated He pneumoperitoneum group and control group ; the expression of E-cadherin in simulated CO2 pneumoperitoneum group was lower than simulated He pneumoperitoneum group and control group;compared with control group, the adhesion and the expression of E-cadherin was not significant difference in simulated He pneumoperitoneum. The weight of metastases in CO2 pneumopefitoneum group was higher than He pneumoperitoneum group and control group; The weight of metastases was not significant difference in He pneumoperitoneum group and control group. Conclusion CO2 pneumoperitoneum may promotes the metastasis of cancer, The influence of He pneumoperitoneum on metastasis of cancer is not significant.
