天花粉蛋白的构效关系及进入细胞的途径 被引量:5

摘要 天花粉蛋白(Trichosanthin,TCS)因具有妊娠引产、抗肿瘤、抗病毒及免疫调节等多种药理学作用而倍受关注。研究天花粉蛋白的结构特征,明确其活性中心及过敏原性区域对于深入了解天花粉蛋白的生物学功能十分重要。该文对近年来关于天花粉蛋白构效关系的研究进展作了综述,并着重介绍了天花粉蛋白进入细胞的具体途径,为研究其他的核糖体失活蛋白提供新的思路。 Trichosanthin (TCS) , a ribosome inactivating protein extracted from the root tuber of Chinese medicinal herb Tri- chosanthes kirilowii Maximowicz, has multiple pharmacological properties including abortifacient, anti - tumor, anti - HIV and immune regulation. The antigenicity and short plasma half - life limited its wide clinical administration. In this review, studies about locating antigenic sites and prolonging plasma half- life were discussed. Studies on structure - function relationship and mechanism of cell entry were also covered. Recently, TCS has been found to induce apoptosis, enhance the action of chemokines and inhibit HIV - 1 intergrase. These findings give new insights on the pharmacological properties of TCS and other members of ribo- some - inactivating proteins.
