摘要
目的 探讨去甲斑蝥素(norcanlharidin,NCTD)对细胞因子诱导的杀伤细胞(cylokine—indueedkillercells,CIK)增殖及CIK细胞杀伤活性的影响。方法采用Ficoll密度梯度法提取脐血单个核细胞,定向诱导成CIK细胞,实验分为加入终浓度为30ixmoL/LNCTD的NCTD—CIK组和不加NCTD的对照CIK组。通过细胞计数法、流式细胞术及MTT法观察CIK细胞的增殖、免疫表型及细胞毒活性的变化。结果NCTD—CIK组细胞增殖率和抗瘤活性均明显高于对照CIK组,差异均有统计学意义(P〈0.05),结论NCTD促进CIK细胞扩增,增强其抗瘤活性,为CIK细胞的过继性免疫治疗提供了一种新的方法。
Objective To explore effects of noreantharidin(NCTD)on the proliferation and killing activity of cytokine - induced killer cells(CIK). Methods Cord blood mononuclear cells were extracted by Ficoll density gradient method and then induced into CIK cells. The experiment was divided into NCTD - CIK group with a final concentration of 30μmol / L NCTD and control group CIK without NCTD. Changes in the proliferation of CIK cells,the immune phenotype and cytotoxic activity were observed using cell counting, flow cytometry and MTT method. Results The rate of cell proliferation and anti - tumor activity in NCTD - CIK group were significantly higher than those of the control CIK group, which showed statistical significance( P 〈 0.05 ). Conclusion NCTD promoted the expansion of CIK cells and enhanced their anti -tumor activity,which will provide a new approach for adoptive immunotherapy of CIK.
