摘要
分别采用荧光、圆二色谱法及活性测定法研究了柚皮苷与α-糜蛋白酶(α-CT)的相互作用。考察了柚皮苷与α-CT的相互作用机理,测定了不同温度下该反应的结合常数及热力学参数。实验结果表明:柚皮苷分子与α-CT作用导致α-CT内源荧光猝灭,猝灭机理主要为静态猝灭,它们之间主要为氢键或范德华作用力;CD光谱研究表明柚皮苷能够引起α-CT的构象变化,使α-螺旋含量减少;酶活性实验表明柚皮苷对α-CT酶活性的影响不大。
The interaction of naringin and α-chymotrypsin (α-CT) were studied by fluorescence and circular dichroism (CD). The results showed that naringin can strongly bind to α-CT,which resulted in the clear fluorescence quenching of α-CT. The Van der waals force and hydrogen-bonding interactions played important roles in binding of naringin to α-CT. On the other hand, the conformation and secondary structure of α-CT were further studied through CD spectra,and the change of α-CT enzyme activity was also determined by uv-vis spectroscopic method. The results indicated that the conformation of α-CT changed after addition of riaringin,α-helix content of α-CT decreased,and the activity of enzyme kept constant.
