摘要
目的研究5-氟尿嘧啶(5-Fu)和顺铂(CDDP)对舌癌细胞的体外杀伤效应,并探讨其与舌癌组织中端粒酶表达的关系。方法将30例舌癌细胞制备成单细胞悬液,分为4组:5-Fu组加入5-Fu;CDDP组加入CDDP;联合用药组加入5-Fu和CDDP;对照组不加任何药物。四甲基偶氮唑盐(MTT法)检测各组细胞生长抑制效果,PCR-ELISA法检测各组端粒酶活性。结果与对照组比较,5-Fu组和CDDP组对舌癌细胞有体外杀伤效应,能抑制舌癌组织端粒酶活性,联合用药组效果更佳(P<0.05或P<0.01)。结论5-Fu和CDDP联合用药对人舌癌细胞的抵制具有协同加强作用,其机制可能是对舌癌细胞的端粒酶活性有较强的抑制作用。
Objective To evaluate antitumor effects of 5-Fu and CDDP drugs in vitro,and investigate their relationship with expression of telomerase in human tongue cancer tissues.MethodsFresh samples of tongue cancer obtained from operation were separately exposed to 5-Fu and CDDP for 24 hour,while the cancer cells in the contrast group were treated without any drugs.Metabplic activity and inhibitory rate of the cells were determined by trypan blue exclusion and MTT assay,Telomerase activity was detected by TRAP-PCR-ELISA.Result 5-Fu and CDDP had vitro antitumor effects on human tongue cancer tissues,and restrain telomerase activity of tongue cancer tissues.The combination of 5-Fu and CDDP may be of most effection(P〈0.05 or P〈0.01).Conclusion 5-Fu and CDDP are concurrently effective on restraining the tongue cancer cell and the reason possibly is they have stronger restrain on activity of telomerase of tongue cancer cell.
