摘要
目的:检测TNF-α、IFN-γ诱导HaCaT细胞β-防御素的表达。方法:用TNF-α、IFN-γ、TNF-α加IFN-γ干预HaCaT细胞后,RT-RCR方法检测hBD-2 mRNA、hBD-3 mRNA的表达。结果:TNF-α干预组HaCaT细胞中hBD-2 mRNA、hBD-3 mRNA,IFN-γ干预组HaCaT细胞中hBD-3 mRNA表达增高(P<0.05),干预12 h时的表达量高于干预24 h时的表达量;TNF-α与IFN-γ联合干预HaCaT细胞12 h时,hBD-2 mRNA、hBD-3 mRNA表达量最高。结论:TNF-α能刺激人角质形成细胞产生hBD-2和hRD-3,而IFN-γ仅能诱导产生hBD-3。
Objective: To detect the expression of human β - defensin - 2 (hBD - 2) mRNA, β - defensin - 3 (hBD - 3) mRNA induced by TNF- α, IFN - γ in human keratinocyte cell lines (HaCaT cells). Methods: Cultured HaCaT cells were stimulated with TNF - α or IFN - γ, or a combination of TNF - α and IFN - γfor 12 h or for 24 h, respectively. The expression of hBD - 2 mRNA, hBD - 3 mRNA was detected by reverse transcription polymerase chain reaction. Results: The expression of both hBD - 2 mRNA and hBD - 3 mRNA was increased in HaCaT cells when induced by TNF - α, and only hBD - 3 mRNA expression was up - regulated when induced by IFN - γ. The expression was stronger at 12 h than at 24 h. 3he expression was strongest when induced by the combination of TNF - α and IFN- γ Conclusion: The expression of both hBD - 2 mRNA and hBD - 3 mRNA was increased in HaCaT cells when induced by TNF - α, and only hBD - 3 mRNA expression np - regulated when induced by IFN - γ.
