摘要 目的:探讨荧光定量聚合酶链反应(FQ-PCR)技术用于孕妇外周血浆中游离胎儿DNA检测的可行性。方法:利用FQ-PCR技术特异性扩增Y基因性别决定区(SRY)并定量分析妊娠中期(18~25周)孕妇血浆中含量。结果:148例男胎孕妇血浆标本中,SRY基因检出率为98.65%。139例女胎孕妇血浆标本中未检测到SRY基因。结论:FQ-PCR技术在孕妇外周血游离胎儿DNA的检测中具有较高的灵敏性和特异性,FQ-PCR技术可应用于非创伤性产前诊断。 Objective: To explore the application value of quantitative fluorescent polymerase chain reaction (FQ- PCR) in the measurement of cell -free fetal DNA in the maternal circulation. Methods: The sex - determining region Y (SRY) gene of cell - free fetal DNA in the maternal ( gestational age of !8 to 25 weeks) circulation was detected with FQ - PCR. Results :The SRY gene was detected in the 98.65% of the samples from 148 women who had the male fetuses. None of the SRY gene was detected in the samples from 139 women who had the female fetuses. Conclusion:The analysis of cell -free fetal DNA -SRY gene in the maternal circulation with FQ - PCR is highly sensitive and specific. It can be developed for non - invasive prenatal diagnosis.
