摘要
目的观察三硝基苯磺酸(TNBS)法溃疡性结肠炎大鼠结肠黏膜TLR-4、NF-κBp65的变化特点及清肠化湿方对TLR-4、NF-κBp65蛋白表达的影响。方法采用TNBS诱导大鼠溃疡性结肠炎模型。将大鼠随机分为正常对照组、模型对照组、清肠化湿方组、巴柳氮组,治疗10d后处死大鼠取新鲜结肠标本,观察黏膜大体形态及组织学改变,并采用Westernblot法检测TLR-4、NF-κBp65蛋白表达水平。结果模型对照组TLR-4、NF-κBp65蛋白表达均高于正常对照组,清肠化湿方组TLR-4、NF-κBp65的表达低于模型对照组,同时清肠化湿方组大鼠结肠形态及组织学损伤评分均有降低。结论清肠化湿方对溃疡性结肠炎模型大鼠具有治疗作用,抑制TLR-4、NF-κBp65的表达可能是其作用机制之一。
Objective: To observe the effect of Qingchanghuashifang Decoction (QD) on the protein expressions and change character of TLR-4 and NF-κBp65 in colonic mucosa of rats with ulcerative colitis (UC). Methods: Rat UC model induced by trinitrobenzene-sulfonic acid (TNBS) were randomly divided into normal group, model group, QD group, and balsalazide sodium tablet (BST) group. The rats were sacrificed to get their colonic mucosa and extract the whole-cell protein after 10d of treatment. Western blot was performed to detect the protein levels of TLR-4 and NF-κBp65. Results: The relative protein expressions of TLR-4 and NF-κBp65 in the model group were all higher than those in the normal group signifieantly. The relative protein expressions of TLR-4 and NF-κBp65 in the QD group were lower than that in the model group. The colonic eonfigurative and histologie injury score were declined. Conclusion: Qingchanghuashifang Decoction can cure ulcerative colitis. One of the mechanisms may be that Qingchanghuashifiang Decoction can inhibit the relative protein expressions of TLR-4 and NF-κBp65.
