Expression and significance of tartrate-resistant acid phosphatase during the stage of physiological root resorption of rabbits

摘要 目的研究家兔下颌第二乳磨牙生理性根吸收时期抗酒石酸酸性磷酸酶(TRAP)在乳牙根周围和恒牙胚周围以及两者之间结缔组织中的表达及其意义。方法分别取0、4、10、16和22d龄乳兔包含乳磨牙牙根及其周围组织在内的部分下颌骨,拍摄其X线片,制备石蜡切片,分别行苏木精-伊红和TRAP染色以及TRAP阳性细胞计数。结果家兔下颌第二乳磨牙牙根于出生后第4天开始吸收;至第10、16天时,根吸收约1/3和2/3;至第22天左右,第二乳磨牙脱落。第0天时,TRAP仅表达于恒牙胚的周围单核细胞。第4天时,乳牙根周围的TRAP阳性单核和多核细胞开始增多,并于第10天达到高峰且差异与第0和4天龄组有统计学意义,第16天时TRAP阳性细胞数开始下降。结论TRAP阳性细胞的位置和水平与乳牙生理性的根吸收进程密切相关,TRAP阳性细胞在恒牙胚周围细胞中出现的规律可能起着重要的作用。 Objective To investigate the function and expression of tartrate-resistant acid phosphatase(TRAP) during the stage of physiological resorption of the second primary molar of rabbits in the roots,surrounding the permanent dental germ and the peripheral tissue.Methods Rabbits of different days of 0,4,10,16,22 were selected as objects.Mandible was used for observing and taking the X-ray,and the roots and peripheral tissue of primary molar teeth for paraffin slicing and hematoxylin and eosin,TRAP dyeing and cell counting.Results The root resorption started on the fourth day and root was absorbed 1/3,2/3 on the 10th and 16th day,to day 22,the shedding occur normally also the permanent tooth erupted.TRAP-positive cells were initially detected in the portion of the dental follicle of the permanent tooth at day 0 postnatal.During 4-10 days postnatal,the number of TRAP-positive mononuclear and multinucleate odontoclasts on the root surface gradually increased but decreased to day 16.Conclusion The regularity in the expression of TRAP during the stage of physiological resorption matched the result of X-ray observation.TRAP-positive cells initially detected in the portion of the dental follicle of the permanent tooth may play an important role in the mechanism of physiological root resorption.
