摘要
目的探讨非小细胞肺癌中ASPP1和ASPP2蛋白表达与细胞凋亡和增殖的关系及其临床意义。方法采用免疫组化Evision法,检测肺癌组织芯片中90例肺癌和25例相应癌旁组织ASPP1、ASPP2及Ki-67的表达;用TUNEL法检测细胞凋亡,以增殖指数和凋亡指数计数表示增殖与其凋亡。结果肺癌组织中ASPP1的表达显著低于癌旁正常肺组织(χ2=5.188,P<0.05),ASPP1的表达与患者的性别、年龄和组织类型无关,而与肺癌分化程度、临床分期和淋巴结转移密切相关(χ2=4.680、6.330、6.045,P<0.05);而ASPP2在肺癌组织及癌旁组织中的表达差异无统计学意义(χ2=0.437,P>0.05);在肺癌组织中细胞凋亡指数(AI)和增殖指数(MI)呈正相关(r=0.622,P<0.05);AI随着ASPP1的表达量增加而增高(t=2.337,P<0.05),而MI与ASPP1的表达无关(t=2.042,P>0.05)。结论肺癌组织中ASPP1低表达及由此诱导的细胞凋亡与增殖失衡可能在肺癌发生、发展中起重要作用。
Objective To investigate the expression of ASPP1 and ASPP2,and to explore its relationship with apoptosis and cell proliferation in non-small cell lung cancer(NSCLC).Methods The expressions of ASPP1,ASPP2 and Ki-67 were determined by immunohistochemical staining combined with tissue micro array of lung cancer,which contained samples of 90 cases of NSCLC and 25 normal adjacent lung tissue.Terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL) was applied to detect the apoptosis in the tissue micro array.The apoptosis index(AI) and mitotic index(MI) were defined.Results The positive rate of ASPP1 was significantly lower in cancer tissues than that in the normal adjacent tissues(χ^2=5.188,P〈0.05).The expression of ASPP1 had close relation with the differentiation,TNM stage and lymphonode metastasis of the disease(χ^2=4.680,6.330,6.045,P0.05),but was not related to sex,age and histological type.There were no significant differences in expression of ASPP2 between lung cancer tissue and the paracarcinomatous tissues(χ^2 =0.437,P〉0.05).The AI and MI had positive correlation in lung cancer tissue(r=0.6220,P〈0.05).The AI increased with the increase of positive ASPP1(t=2.337,P〈0.05),but MI had no relationship with ASPP1(t=2.042,P〈0.05).Conclusion The lower expression of ASPP1 in lung carcinomas and inducing apoptosis and disequilibrium of proliferation may be play important roles in the early stages of the tumor genesis.
