钙调神经磷酸酶及GATA-6在大鼠主动脉钙化中的作用

摘要 目的观察钙调神经磷酸酶(calcineurin,CaN)和锌指转录因子6(zinc finger GATA transcription-6,GATA-6)在大鼠主动脉钙化中的作用。方法用华法令和维生素D_3建立大鼠血管钙化模型,4周龄雄性SD大鼠36只随机分成对照组,钙化组和钙化+环孢霉素A(cyclosporin A,CsA)组。HE染色及电镜下观察血管结构变化;原位杂交方法检测各组GATA-6 mRNA及CaNAαmRNA的表达;免疫组织化学方法检测各组GATA-6及CaNB1蛋白表达,测定主动脉组织CaN活性和碱性磷酸酶活性。结果钙化组和CsA组GATA-6 mRNA、蛋白及CaNAα mRNA、CaNB1蛋白的表达增加(P<0.01),钙化组和CsA组CaN活性及ALP活性增加(P<0.01)。结论 CaN和GATA-6参与血管钙化调节,CsA可能促进血管钙化。
