摘要
目的探讨活血抗生滴丸对佐剂型关节炎(AA)大鼠炎症和免疫功能的影响。方法建立Wistar大鼠AA模型,随机分为5组:活血抗生滴丸0.63、1.25、2.5g/(kgd)剂量组、环孢素A10mg/(kgd)组及模型组。于造模的第19天开始给药,1次/d,连续灌胃30d。检测各组大鼠关节肿胀度以及脾脏、胸腺指数和外周血中免疫球蛋白IgG、IgM、IgA及T淋巴细胞亚群含量。结果活血抗生滴丸2.5g/(kgd)组和环孢素A组能明显降低AA大鼠关节肿胀度,降低脾脏、胸腺指数,降低外周血中IgG含量,减少CD4+细胞数,升高CD8+细胞数,降低CD4+/CD8+比值(P<0.01)。结论活血抗生滴丸可抑制AA大鼠炎症发展,对其体液免疫及细胞免疫均有一定抑制作用。
Objective To investigate the effects of Huoxuekangsheng dripping pills on the inflammation and immunological function of rats with adjuvant-induced arthritis(AA). Methods Wistar rats with established adjuvant-induced arthritis were divided at random into five groups were treated with Huoxuekangsheng dripping pills 0.63, 1.25, 2.5g/(kg-d) and cyclosporine A(CsA) 10mg/(kg.d) on the 19th day, ig, dayly, for 30 days . The effects of treatment were valuated by measurement of the swell in joints, spleen index, thymus index. The peripheral blood immune globulin IgG, IgA, IgM were measured by single radical immunodiffusion test and the T lymphocyte subsets CD4^+, CD8^+ and the ratio of CD4^+/CD8^+ were detected by flow cytometry. Results Compared with AA models, groups of Huoxuekangsheng dripping pills 2.5g/(kg.d) and CsA could remarkably ameliorate the swell in joints, spleen and thymus index, also the level of IgG, CD4^+ are significantly decreased, but CD8^+ is increased, so the ratio of CD4^+/CD8^+ is decreased(P〈0.01). Conclusion Huoxuekangsheng dripping pills can attenuate the degree of inflammation of AA rats and its mechanism might be associated with its inhibition of the humoral immunity and cellular immunity function.
