摘要
分别以普通饲料+STZ,普通饲料+柠檬酸对照,高脂饲料+STZ和高脂饲料+柠檬酸对照4种方式处理购自北京、南京和上海共4个生产单位的雄性C57BL/6J(B6)小鼠.分别于注射(STZ或柠檬酸对照)前和注射后3周连续测定非空腹血糖浓度.结果显示虽然高脂饲料联合注射STZ的方法可以较好模拟2型糖尿病血糖变化,但是B6小鼠的遗传背景(来源)也是影响模型成败的关键因素之一.
Four populations of C57BL/6J (B6) strain purchased from Beiiing, Naniing and Shanghai were sampled randomly and weaned onto standard rodent chow and high-fat diet (HFD) respectively. After exposure to the diets for five weeks, the experimental groups were injected intraperitoneally with streptozo- tocin, while the control groups were injected with vehicle. All the mice were kept on the same diet for next three weeks, and non-fasting blood glucose levels were measured before and after STZ injections. The present study indicates that HFD combined with subsequent injection of STZ is valid to stimulate the chan- ges of blood glucose in type 2 diabetes, and that the inconsistence compared with previous studies may be attributed to genetic variation of the B6 mice maintained in different commercial breeders.
