应用珊瑚作为重组人骨形成蛋白-2缓释载体的动物实验研究 被引量:2

摘要 将大肠杆菌表达的重组人骨形成蛋白-2(rhBMP-2)和珊瑚(coral)复合,制成rhBMP-2/coral复合人工骨,将其植入小鼠股部肌肉内,并以单纯珊瑚植入作对照,术后1、3、6周取材,组织学观察。结果显示:rhBMP-2/coral复合人工骨在植入局部诱导形成骨和软骨,表现出良好的异位诱导成骨能力;珊瑚对rhBMP-2的活性未产生不利影响,是rhBMP-2的良好缓释载体。此复合人工骨可望作为一种新型生物性植骨材料而应用于骨科和颌面外科。 By combining coral with recombinant human bone morphogenetic protein-2 (rhBMP-2), rhBMP-2/coral composited artificial bone was obtained; then, the composite was implanted into the muscle pouches of mice, coralused as control. The mice were sacrificed at 1, 3, 6 week respectively after operation and ectopic osteoinductive activity was examined by histology. The results showed that the composite possessed definitive osteoinductivity andinduced endochondral bone formation in the regions of implantation; coral did not disturb the osteoinductive activity of rhBMP-2 and acted as one of the most suitable rhBMP-2 slow-release carriers currectly available. Thecomposite will be a new type of bone substitute in orthopaedics and maxillofacial surgery.
