纳米TiO_2影响细胞生长活性的尺寸效应

The size-effects of nano-TiO_2 on the viability of cultured cell

摘要 目的不同尺寸纳米TiO2(5、10、15和60 nm)暴露于不同组织来源细胞(HPF、ESF、U251、HB),探讨纳米TiO2对细胞生长活性的影响,探讨不同尺寸、不同浓度纳米TiO2粒子与细胞生长之间的相关性,分析纳米TiO2的细胞生物学效应。方法不同尺寸、不同浓度纳米TiO2暴露不同组织来源的体外培养细胞,使用MTT方法检测细胞生长活性,计算细胞半致死量,并运用SPSS10.0软件进行统计学分析。结果10 nm以上的TiO2粒子暴露能显著抑制肺来源(HPF)和神经来源的细胞(U251)生长活性,对皮肤(ESF)和膀胱组织(HB)来源细胞生长活性抑制较弱;5 nmTiO2颗粒对细胞生长活性基本无抑制作用,相反,还观察到对HPF、ESF、HB细胞有增强细胞生长的情况。结论在本试验条件下,纳米TiO2对细胞生长活性的影响有尺寸效应,纳米TiO2暴露所导致的生物学效应有细胞间差异,其对HPF、U251细胞影响较明显,对ESF、HB影响较弱。 Objective To investigate the growth activity and biological effects of different sources cells(HPF,ESF,U251,HB),which were exposured to the different sizes Nano TiO2(5,10,15,60 nm),and the correlation between different sizes of nano-particles and cell growth.Methods exposuring the cultured cells in vitro derived from different tissues to different sizes and different concentrations of Nano TiO2 particles,detecting the cell growth activity using MTT technique,calculating the semi-lethally,and using SPSS 10.0 software for statistical analysis.Results The TiO2 particles exceeding 10 nm may significantly inhibit cell growth activity of HPF and U251,and less inhibit growth activity of ESF and HB cell.The 5 nm TiO2 particles had little inhibition on cell growth activity,more over,may enhance cell growth activities of HPF,ESF,HB.Conclusion there were size-effects of Nano TiO2 on cell growth activities,which was more significant in HPF and U251 than in ESF and HB.
