摘要
采用热水浸提法,以粗多糖得率作为评价指标,通过单因素试验和正交试验,对花脸香蘑[Lepista sordida(Fr.)Sing.]菌丝体发酵全液中多糖的提取条件进行优化。结果表明,优化的提取条件为:提取温度100℃、提取次数3次、pH 12、乙醇终浓度70%;在此优化条件下,花脸香蘑发酵全液的粗多糖得率达4 mg/mL。
Orthogonal testing [L9(34)] was used to define more precisely the factors,determined on the basis of single factor experiments,affecting crude polysaccharide extraction from submerged cultures of Lepista sordida.Data revealed that the optimal extraction conditions consisted of three sequential aqueous extractions conducted at pH 12 and 100 ℃ for 1 h,followed by precipitation with ethanol at a final concentration of 70%.Under these conditions,the yield of crude polysaccharide extracted was 4 mg/mL.
