川芎嗪对肺癌A549细胞增殖与凋亡的影响及其机制 被引量:15

Effect of Tetramethypyrazine on the Proliferation and Apoptosis of Lung Cancer Cell Line of A549 and Its Mechanism

摘要 目的:研究川芎嗪对肺癌A549细胞增殖与凋亡的影响,并探讨川芎嗪治疗肺癌的作用机制。方法:选用肺癌A549细胞体外培养,在不同浓度川芎嗪作用下,应用MTT法检测肺癌细胞的增殖抑制,应用流式细胞仪检测肺癌细胞的凋亡及细胞周期的分布,采用免疫细胞化学方法检测川芎嗪对肺癌A549细胞Bcl-2及Bax表达的影响。结果:川芎嗪对肺癌A549细胞增殖有明显抑制作用,且呈时间剂量依赖关系;川芎嗪能够使A549细胞G0/G1期、G2/M期细胞增多,与对照组比,差异显著(P<0.01);川芎嗪能够下调肺癌A549细胞Bcl-2的表达,对Bax无明显影响,使Bcl-2/Bax下降。结论:川芎嗪对肺癌A549细胞的增殖有抑制作用,其机制可能与川芎嗪能够改变A549细胞周期分布,下调肺癌A549细胞Bcl-2表达,改变Bcl-2/Bax比例,促进肿瘤细胞凋亡等有关。 Objective: To investigate the inhibitory effect of tetramethypyrazine (TMP) on the proliferation and apoptosis of the lung adenocarcinoma A549 cell line and to explore its mechanism in the treatment of lung cancer. Methods: Different doses of TMP were added to the lung adenocarcino ma A549 cell line to demonstrate the biological function. MTT was performed to reveal the inhibitory effect on cell proliferation, and flow cytometry was used to determine the apoptosis and cell cycle of the lung adenocarcinoma A549 cell line after the treatment of TMP. Bcl-2 and Bax were measured by immunocytochemistry. Results: There was a time- and dose-dependent inhibition of cell proliferation of A549 by TMP. TMP increased the rate of G0/G1 and G2/M of A549 cell cycle, down-regulate the expression of Bcl-2, and decrease the rate of Bcl 2/Bax so as to promote the apoptosis of A549. Conclusion: TMP can inhibit the proliferation of lung adenocarcinoma A549 cell line through changing cell cycle, down-regulating Bcl-2 expression, and decreasing the rate of Bcl-2/Bax.
