摘要
目的探讨上矢状窦闭塞后脑组织中水通道蛋白-4(AQP-4)表达与血-脑脊液屏障破坏之间的关系。方法选择健康雄性Wistar大鼠49只,分为对照组和实验3h、6h、12h、24h、3d、7d组,每组7只。采用结扎法建立大鼠上矢状窦闭塞模型,对其脑组织含水量、血-脑脊液屏障破坏情况进行观察,用免疫组化法检测AQP-4蛋白的表达。结果脑组织含水量在上矢状窦闭塞6h开始增加,持续升高3d时达到高峰,实验6h、12h、24h、3d、7d组与对照组比较,差异均有统计学意义(P<0.05)。与对照组相比,实验组脑组织伊文氏蓝(EB)含量6h、12h、24h、3d明显升高,差异均有统计学意义(P<0.05),24h时EB含量达到高峰。AQP-4表达在上矢状窦闭塞后6h开始上调,24h达高峰,维持3d时开始逐渐回落,除3h外,实验组各时相点相同部位AQP-4的表达与对照组比较,差异均有统计学意义(P<0.05)。AQP-4表达与脑组织含水量的相关性分析表明,两者呈正相关(r=0.846,P<0.01)。结论上矢状窦闭塞后脑组织含水量显著增加,AQP-4增高在血-脑脊液屏障破坏及血管源性脑水肿形成过程中发挥了重要作用。
Objective To investigate the relationship between Aquaporin - 4 ( AQP - 4) expression and blood - brain barrier damage after superior sagittal sinus obstruction in rats. Methods Forty - nine healthy male Wistar rats were divided into groups control, experimental 3h, 6h, 12h, 24h, 3d, 7d, 7 in each. Superior sagittal sinus occlusion models were established by ligation. Brain tissue water content and blood - brain barrier damage were observed, and AQP - 4 protein expression deter- mined by immunohistochemistry. Results Brain tissue water content began to increase at hours 6 after superior sagittal sinus occlusion, peaking after 3 days of continuous rise, and experimental groups 6h, 12h, 24h, 3d, 7d were significantly different from control (P 〈0. 05). When compared with control group, EB contents at hours 6, 12, 24, days 3 increased in experi- mental groups, the difference was significant (P 〈 0. 05 ). EB peaked at hours 24. AQP -4 expression began to be up -regulated at hours 6, peaking at hours 24, and started to fall after 3 h. There was significant difference in AQP - 4 expression in the same parts at various time points between control and all experimental groups except group 3 h ( P 〈 0. 05 ). Correlation analysis showed that AQP - 4 was positively correlated with brain tissue water content ( r = O. 846, P 〈0. 01 ). Conclusion Increased brain water content and AQP -4 after superior sagittal sinus ocelsion barrier damage play an important role in the blood - brain barrier damage and the formation of vasogenic brain edema.
