摘要
目的通过1型糖尿病(T1DM)大鼠胰岛素(insulin,Ins)水平变化与胰岛β-细胞中端粒酶逆转录酶(TERT)表达及端粒长度关系的分析,探讨TERT及端粒长度变化在T1DM发病机制中的作用。方法T1DM大鼠模型组与正常对照组各48只,分别测定Ins,同时分离胰岛β-细胞,原位杂交法检测胰岛β-细胞TERT的表达,用Southern Blot法检测端粒长度,并进行对比研究。结果T1DM组Ins水平显著低于对照组(P<0.01),T1DM组β-细胞中TERT的表达显著高于对照组(P<0.01),而端粒长度显著低于对照组(P<0.05);T1DM组的Ins水平下降与TERT活性表达增强呈高度负相关(r=-0.8942,P<0.01),与端粒缩短呈正相关(r=0.9238,P<0.01)。结论T1DM大鼠Ins水平降低与TERT活性增强、胰岛β-细胞端粒缩短密切相关。提示胰岛β-细胞中TERT的活性增强,可能是抑制胰岛β-细胞对Ins分泌的病理机制,TERT活性表达及端粒缩短也可作为T1DM病情监测的指标。
[Objective] To investigate the relation between the change's of TERT and length of telomere with pathogenesis of T1DM through analysis of the change of Ins level and expression of TERT and telomere length of T1DM. [Method] T1DM rat group and control one was 48 respectively, they were determined separately Ins and separate pancreatic island β-cell, and were detected the expression of pancreatic island β-cell TERT through ISH, telomere length through southern blot, then we contracted the results of two groups'. [ Results ] The Ins level of T1DM group is lower than control one (P 〈0.01) significantly, expression of TERT of pancreatic island β-eell of T1DM group is significant higher than control one (P 〈0.01), but telomere length of T1DM group is significant lower than control one (P 〈0.05). T1DM group shows high negative correlation between declination of Ins level and strengthen of expression of TERT activity (r =0.9238,P 〈0.01). [Conclusion] The declination of Ins level of T1DM shows directly bound up with the strengthen of TERT activity and reduction of pancreatic island β-cell telomere. The article pointed out that the strengthen of TERT activity may inhibit the Ins secretion of pancreatic island β-cell. As index, the expression of TERT activity and declination of telomere can indicate the disease process of T1DM.
