地黄寡糖及其主要成分对大鼠脑片氧化应激损伤保护作用研究 被引量:10

摘要 目的:探讨地黄寡糖及其主要组分对H2O2所致大鼠大脑皮质和海马脑片氧化应激损伤的保护效果。方法:取SD大鼠大脑皮质和海马制作脑片,然后随机分为对照组(Control);单纯地黄寡糖用药组(ROS);单纯水苏糖组(stachyose);单纯梓醇组(catalpol);H2O2损伤组(H2O2);损伤前地黄寡糖预处理组(ROS+H2O2);损伤前水苏糖预处理组(stachyose+H2O2);损伤前梓醇预处理组(catalpol+H2O2)。TTC染色法检测脑片组织损伤百分率,比色法检测培养液中乳酸脱氢酶(LDH)含量。结果:H2O2孵育30min明显损伤大鼠海马和皮质脑片,组织损伤率明显升高,LDH释放增加(P<0.01)。8、40、200 mg/L的地黄寡糖或120 mg/L的水苏糖、20 mg/L的梓醇可不同程度改善H2O2所致的氧化应激损伤引起的组织损伤,减少LDH的漏出,但各成分均对对照组脑片无影响。结论:地黄寡糖、水苏糖、梓醇均能减轻H2O2所造成的氧化应激损伤。梓醇相比水苏糖在地黄寡糖中起主要作用。 Aim:To investigate the protection of Rehmannia glutinosa oligosaccharides(ROS) and its components against oxidative stress injuries induced by H2O2 on rat cerebral cortical and hippocampal slices,and compare which is more effective between stachyose and catalpol.Method: Rat cerebral cortex and hippocampus slices were made and divided eight groups,randomly:(Ⅰ)normal cultures;(Ⅱ)ROS control cultures;(Ⅲ) stachyose control cultures;(Ⅳ) catalpol control cultures;(Ⅴ) H2O2-exposed control cultures;(Ⅵ) H2O2-exposed control cultures pretreated with ROS;(Ⅶ) H2O2-exposed control cultures pretreated with stachyose;(Ⅷ) H2O2-exposed control cultures pretreated with catalpol.Then the rate of tissue injury was determined by TTC staining,and lactate dehydrogenases(LDH) were detected by colorimetric method.Result: Incubated with 2mmol/L H2O2 for 30min,rat cerebral cortical and hippocampal slices were significantly damaged,indicated by decreased A490nm value of TTC staining.Meanwhile the release of LDH in supernatant increased.ROS(8,40,200 mg/L),stachyose(120 mg/L),catalpol(20 mg/L) pre-incubation for 30min significantly inhibited the decrease in TTC value and the elevation of LDH release in different degree,which depended on doses in certain range.Conclusion: ROS,stachyose,catalpol can protect rat cerebral cortical and hippocampal slices against injury induced by H2O2.Comparing with stachyose,the catalpol is more effective in ROS.
