摘要
目的观察泡球蚴感染BALB/c小鼠肝脏转化生长因子β1(TGF-β1)的动态变化。方法将60只BALB/c小鼠随机分为实验组和对照组,肉眼直视下肝脏穿刺感染泡球蚴。分别于感染后2 d8、d、4周、12周、24周、36周采取小鼠肝脏组织,通过HE和Masson染色观察小鼠肝脏病理改变,免疫组织化学法观察泡球蚴感染不同阶段的TGF-β1动态变化。结果泡球蚴感染小鼠肝脏汇管周围出现炎细胞浸润、脂肪变性、坏死、钙化和肝泡球蚴纤维囊壁形成等多种病理改变;TGF-β1表达水平呈先升高再下降趋势,感染4周、12周、24周、36周时的阳性率分别为(1.44±3.22)%、(18.83±4.03)%(、9.44±4.71)%和(8.13±3.65)%,其中感染12周和36周与对照组相比差异有统计学意义(P<0.05)。结论泡球蚴感染BALB/c小鼠TGF-β1表达上调,可能与感染小鼠肝纤维化的发展有关。
Objective To observe the dynamic expression of TGF-β1 and pathological features of the livers of BALB/c mice infected with Echinococcus multilocularis(Em).Methods Sixty BALB/c mice were randomly divided into 2 groups,an experimental group and a control group.Em infection was performed after laparotomy by intra-hepatic injection of a metacestode suspension(experimental animals) or of a medium used to prepare the metacestode suspension(control animals) under a surgical microscope.After 2 days,8 days,4 weeks,12 weeks,24 weeks and 36 weeks,samples of mouse liver were collected and subjected to HE and Masson staining.The dynamic changes in liver TGF-β1 expression in BALB/c mice infected with Em was analyzed immuno-histochemically.Results At different times during the course of Em infection,pathological changes were observed in the liver of the experimental group,including inflammatory cell invasion,steatosis,necrosis,calcification,and the formation of cyst walls.The level of TGF-β1 expressi...更多on increased as the hepatic tissue morphology changed to hepatocytic stroma.In the experimental group,the expression of TGF-β1 was positive in(1.44±3.22)%(4 weeks),(18.83±4.03)%(12 weeks),(9.44±4.71)%(24 weeks),and(8.13±3.65)%(36 weeks);in the control group,this expression was positive in(2.43±1.91)%(24 weeks) and(0.29±0.65)%(36 weeks).There was a significant difference between the experimental group and the control group at 12 weeks and 36 weeks(P〈0.05).Conclusion TGF-β1 expression in the liver of mice infected with Em increased and remained high during Em infection and may have been involved in the pathogenesis and development of liver fibrosis.
