纤维素酶-超声辅助提取麦麸总黄酮的工艺研究 被引量:12

Study on total flavones extraction from wheat bran under the intergrated treatment with ultrasonic and cellulase

摘要 以小麦麸皮为原料研究纤维素酶与超声辅助提取黄酮类物质的作用规律,考察反应条件对总黄酮提取率的影响,对不同提取方法的提取效果和粗提物的DPPH清除活性进行了比较,并采用正交试验对提取工艺进行了优化研究。结果表明最佳提取条件为:乙醇体积分数60%、温度50℃、pH5.5、加酶量0.55%、料液比1∶25、超声功率60W、提取时间30min,确定提取级数为2次。在最佳工艺参数下提取率可达0.2355%。采用纤维素酶与超声辅助提取,可有效提高麦麸总黄酮的提取率,粗提物得率高,且DPPH清除活性较强。 Wheat bran was treated in cellulose and ultrasonic,the extraction rate of total flavones was measured under different treatment condition. The extraction effects of different methods and DPPH radical scavenging activity of crude extracts were compared. The optimum condition of extraction was obtained through orthogonal test. Experiment results show that the optimum extraction parameters were:60% volume fraction of ethanol,temperature of 50 ℃,pH5.5,0.55% dry weight of cellulase,solid to liquid ratio of 1 ∶25,ultrasonic power of 60 W, extraction time of 30 min, extraction series of 2. Under such condition the extraction rate of total flavones was up to 0.2355%. The cellulose-ultrasonic extraction method shows high effectiveness and the crude extract has the highest yield with stronger DPPH radical scavenging activity.
