摘要
目的:探讨葱白提取物(FOB)抗大鼠离体心脏缺血再灌注(I/R)损伤和减少心肌细胞内Ca2+的增加。方法:利用Langendorff心脏灌流系统建立大鼠离体心脏I/R模型,采用激光扫描共聚焦显微镜,借用Fluo-3/AM荧光染色技术观察心肌细胞内Ca2+强度。结果:①FOB对大鼠离体心脏I/R损伤具有明显的保护作用,随FOB剂量的增加,使左室发展压(LVDP)的恢复更加接近缺血之前的水平,再灌注痉挛度更加减轻,心脏从缺血后恢复跳动更迅速,心律失常的发生越来越少,与I/R组相比,100g/LFOB使LVDP的恢复从(20.3±2.8)%增加到(94.8±6.4)%(P<0.01),再灌注痉挛度从(89.2±9.7)mmHg减轻为(10.2±3.1)mmHg(P<0.01)。②FOB使I/R损伤后心室肌细胞内Ca2+染色荧光强度明显减少,随着FOB浓度增高,减少细胞内Ca2+的作用更加强烈。与I/R组相比,100g/L的FOB使心肌细胞内Ca2+染色荧光强度从(144.8±16.4)减少到了(50.8±11.61)(P<0.01),表明FOB具有减少I/R中细胞内钙超载作用。结论:FOB能抗大鼠离体心脏I/R损伤,可能原因是FOB减轻I/R后心肌细胞内游离钙浓度的增加。
Objective:To investigate the protective effect offistular onion bulb extract(FOB) preconditioning on ischemia-reperfusion(I/R) injury in vitro and and its inhibitory effect on free Ca2+ increase ofcardiomyocytes ofrats.Methods:The hearts models ofI/R in vitro in rats were set up through using Langendorffperfusion system.The fluorescence intensity ofintracellular Ca2+ in cardiomyocytes ofrats was detected with Flup-3/AM fluorescein stain by the laser scanning confocal microscopy.Results:①FOB markedly antagonized I/R injury with dose-dependent in hearts ofrats in vitro.FOB could significantly recover the left ventricular developed pressure(LVDP) ofI/R, relax the magnitude ofreperfusion contracture, and improve the supply offlowing solution ofcoronary artery, and decrease the incidence ofthe cardiarrhythmia.Compared with I/R groups, FOB(100g/L) significantly recovered the LVDP from(20.3±2.8)% to(94.8±6.4)%(P〈0.01),and decreased the reperfusion contracture from(89.2 ±9.7)mmHg to(10.2±3.1)mmHg(P〈0.01).② FOB obviously decreased the free Ca^2+ overloading ofcardiomyocytes ofI/R injury.With concentration-dependent ofFOB, the effects ofFOB reduced Ca^2+ overloading increasingly became strong.Compared with I/R group, FOB significantly decreased the fluorescence intensity ofintracellular Ca^2+ in cardiomyocytes ofrats from(144.8± 16.4)to(50.8±11.61)(P〈0.01).Conclusion:FOB could protect the I/R injury ofhearts ofrats in vitro, which decreased the Ca^2+ overloading ofcardiomyocytes period I/R.
