摘要
目的:观察复方通络胶囊对缺氧-复氧所致人脐静脉内皮细胞(HUVECs)损伤的作用机制。方法:体外培养HUVECs,建立缺氧-复氧模型,收集培养液,测定内皮素(ET)、一氧化氮(NO)、一氧化氮合酶(NOS)水平。结果:复方通络胶囊中、高剂量组能抑制缺氧-复氧诱导的ET释放,提高NOS的活性,促进NO的释放(P<0.05,P<0.01)。结论:复方通络胶囊通过抑制ET释放,提高NOS的活性,促进NO的释放,对人脐静脉内皮细胞缺氧-复氧损伤有明显的保护作用。
Objective:To observe the mechanism of protective effect of Fufangtongluo Capsules on human umbilical vein endothelial cells (HUVECs) injury caused by hypoxia-reoxygenation. Methods:HUVECs were cultured in vitro to build the hypoxia-reoxygenation model,then we collected culture medium to determinate endothelin (ET),nitric oxide (NO),nitric oxide synthase (NOS) level. Results:We observed Fufangtongluo Capsules could inhibit the hypoxia-reoxygenation-induced ET release,improved NOS activity and promoted NO released in medium and high dose groups (P0.05,P0.01). Conclusion:Fufangtongluo Capsules has obviously protective effect on human umbilical vein endothelial cells injury by inhibiting the release of ET,enhancing NOS activity and promoting the release of NO.
