摘要
目的:本研究拟探讨食管癌相关基因2(ECRG2)蛋白产生后在食管癌EC109细胞的分布。方法:利用35S-蛋氨酸标记的Pulse-chase实验示踪ECRG2蛋白产生后在EC109细胞内外的分布情况;利用免疫荧光染色方法观察ECRG2蛋白在EC109细胞内的亚分布。结果:Pulse-chase示踪实验显示ECRG2蛋白产生后,一部分聚集于细胞核内,另一部分被分泌到细胞外;进一步免疫荧光染色分析显示,在间期细胞中ECRG2聚集于中心体处,在有丝分裂期细胞聚集于着丝粒处。结论:ECRG2基因有可能分别在细胞内、细胞外发挥不同的生物学功能,通过细胞外发挥丝氨酸蛋白酶抑制剂功能、细胞内参与中心体复制、纺锤体检测点功能的调控,而在肿瘤细胞侵袭迁移、染色体不稳定性中发挥重要作用。
Objective:To study the distribution of ECRG2 protein in the EC109 cells.Methods:Using pulse-labeled with / methionine and Western blotting to examine the distribution of ECRG2 protein in EC109 cells.Using immunofluoscence staining to observe the localization of ECRG2 in EC109 cells.Results:Pulse-chase experiments showed that ECRG2 accumulated in both supernatant and nucleus.Further immunofluoscence staining showed that ECRG2 localized in centrosomes during interphase and kinetochores in mitosis.Conclusion:ECRG2 protein was transferred to both intracellular and extracellular cells,and may involve in regulation of tumor migration/invasion,centrosome duplification and mitotic spindle checkpoint,and play an important role through different mechanisms during migration/invasion and chromosome instability of cancer.
