摘要
目的:建立测定胰岛素增敏剂罗格列酮游离碱电离平衡常数值的方法并测定罗格列酮游离碱的电离平衡常数。方法:将pH1.64、5.76、6.41、6.91、7.14、7.46、10.64的罗格列酮游离碱甲醇溶液分别在200~400 nm处扫描,并在选定波长312、317、326、330 nm处测定其吸收度,得到罗格列酮游离碱电离平衡常数值。结果:罗格列酮游离碱的pKa值为7.09。结论:该方法简便易行,适用于测定酸碱pKa值在2~9之间的药物。
Objective:To establish a method for determination of ionization equilibrium constant of insulin-sensitizer rosiglitazone free base. Methods: Methanol solutions of Rosiglitazone free base with pH values of 1.64,5.76,6.41,6.91,7.14,7.46 and 10.64 were respectively scanned at the wavelength within the range of 200-400 nm. The absorbance and pKa value of rosiglitazone were determined at the selected wavelength of 312,317,326 and 330nm respectively. Results: The ionization equilibrium constant value of rosiglitazone free base was 7.09.Conclusion : The present method is convenien't and applicable for determination of pKa 2-9 of drugs.
