摘要
目的:观察紫杉醇(PA)抑制人胃癌SGC7901细胞增殖机制影响的实验研究。方法:采用MTT法、集落形成实验、流式细胞仪(FCM)和检测端粒酶活性来观察PA作用于人胃癌SGC7901细胞48h后对肿瘤细胞增殖机制、细胞生长抑制率和细胞周期的变化。结果:0.4、1.0、2.0、4.0、8.0、16.0、32.0、64.0μg/mL的PA对人胃癌SGC7901细胞生长抑制率分别为10.9%、12.7%、24.1%、30.2%、32.5%、45.6%、59.3%和73.4%,PA处理后SGC7901细胞集落形成明显受到抑制,使细胞周期阻滞于S期,端粒酶活性水平下降。结论:PA对人胃癌SGC7901细胞的增殖抑制作用明显。
Objective : To study the mechanisms of paclitaxel which inhibition proliferation of the human gastric carcinoma cell line SGC7901 cells. Methods:By MTT assay and flow cytometry (FCM) were respectively used to determine the influence of paclitaxel which affected the gastric carcinoma cell line SGC7901 cells in its proliferation and cell cycle for 48 hours. Results :The inhibitive rates of gastric carcinoma celt line SGC7901 were 10.9% ,12.7% ,24. l% ,30.2% ,32. 5%, 45.6% ,59.3% and 73.4% with different concentration of Paclitaxel which was 0. 4, 1.0,2.0,4. 0,8.0,16. 0,32.0, 64. 01μg,/mL respectively. The clone formation of SGC7901 cell was evidently inhibited and cell cycle was arrested in S - phase and inhibited telomerase activity after it was treated with paclitaxel. Conclusion :The effect of paclitaxel which inhibited the proliferation of the human gastric carcinoma cell line SGC7901 was related with S - phase arresting of cell cycle.
