摘要
目的:考察辐射剂量对石杉碱甲药剂学性质的影响。方法:石杉碱甲微球采用^(60)Co照射γ射线灭菌,采用高效液相色谱测定石杉碱甲微球的含量,通过透析释药法测定微球的体外释放度。采用扫描电镜观察微球的外观形态。结果:甘露醇按照其和微球0.5:1的比例加入,可以防止微球黏连,10,15,20 kGy的辐射剂量对微球的外观、含量及释放度无明显影响。结论:石杉碱甲微球采用辐射灭菌的方法可行。
OBJECTIVE To evaluate the effects of radio sterilization on the pharmacy characteristics of huperzine A inject able microspbercs. METHODS ^60 Co γ-ray was applied in huperzine A microspheres sterilization. Dialysis release method was used to determine in vitro release rate of huperzine A microspheres. HPLC was used to determine the content of huperzine A microspheres. The morphology of the microspheres was observed by scanning electron microscopy. RESULTS The micro spheres adhension could be avoided after radio sterilization when mannitol was added. Radiation dose (10, 15,20 KGY) had little effects on appearance, drug content and in vitro release rate of microspheres. CONCLUSION ^60Co γ-ray could be used to the sterilization of huperzine A microspheres.
