摘要
目的:建立高效液相色谱—荧光检测法测定盐酸莫西沙星血药浓度的方法。方法:采用乙睛—缓冲液(18:82)为流动相,缓冲液为磷酸二氢钠与四丁基硫酸氢胺混合水溶液;色谱柱:Kromasil C18(250mm×4.6mm,5μm);流速1.0ml·min-l;进样量20μl;柱温:35℃。以环丙沙星为内标,采用荧光检测器检测,激发波长为290nm,发射波长为488nm。结果:盐酸莫西沙星在0.25~5.0μg·ml-1血药浓度范围内线性关系良好(r=0.9997)。结论:该方法操作简便、专属性强、灵敏度高、结果可靠,可用于盐酸莫西沙星血药浓度的测定。
Objective: To develop a HPLC method for determination of Moxifloxacin Hydrochloride in human plasma. Methods: HPLC analysis was performed on Kromasil C18(250 mm×4.6 mm, 5 μm)with the mixture of acetonitrile-buffer solution( 18:82, the buffer solution contain sodium dihydrogen phosphate and tetrabutylammonium hydrogen sulfate), 20 pd of supernatant was injected with the temperature of 35 ℃. Flow-rate was 1.0 ml ·min^-1. The internal standard was ciprofloxacin.The excitation and emission wavelength of fluorescence detector were 290 nm and 488 nm. Results:A good linear relationship was obtained between 0.25-5.0μg ·ml^-1 (r=0.999 7). Conclusion: The method is steady and accurate with a good repeatability and can be used for blood concentration monitoring of Moxifloxacin Hydrochloride.
