聚乳酸生物材料与人乳牙牙髓干细胞的生物相容性研究 被引量:5

摘要 目的:观察聚乳酸生物材料(PLA)与人乳牙牙髓干细胞(hSHED)的生物相容性及其作为支架材料的矿化诱导作用。方法:酶消化法体外培养hSHED,并用免疫组织化学方法鉴定;分别用MTT法和茜素红染色法观察PLA生物材料对hSHED增值活性和矿化能力的影响。结果:成功分离了人乳牙牙髓干细胞,波形蛋白染色阳性,牙本质涎蛋白(DSP)染色阴性,确定为间充质来源的未分化乳牙牙髓干细胞;MTT法检测hSHED在生物材料上的增殖状况与正常培养基本一致;茜素红染色结果显示:浸提液培养组的矿化结节量明显多于空白对照组。结论:聚乳酸生物材料与人乳牙牙髓干细胞有良好的生物相容性,作为支架材料对于乳牙牙髓干细胞有较好矿化诱导作用。 AIM:To observe the adhesion and growth of hSHED (stem cells from human exfoliated deciduous teeth) in the leaching liquor of PLA in vitro and the osteogenic induction of PLA. METHODS : The hSHEDs were cultured, and the cells were identified by immunohistochemical method. The compatibility of the biomaterial was evaluated by MTT assay. Alizarin red staining was employed to measure the osteogenic differentiation of SHED. RESULTS: SHED was successfully isolated and cultured. By immunohistochemical staining, we found that SHEDs were positive for vimentin but negative for DSP. The proliferative ability of SHED on PLA has no obvious difference from the control group by MTT method. The Alizarin red staining showed that calcium deposition in the experiment group was more than that in the control group. CONCLUSION: PLA possesses a good cellular compatibility with SHED. As an engineering scaffold, it induces the osteogenic differentiation of SHED.
