摘要
报道了L谷酰胺产生菌Bc-66诱变育种及其摇瓶发酵条件研究的结果。以黄色短杆菌ATCC14067为出发菌种,经逐级诱变处理,选育1株能高产L谷酰胺,副产物较少,且对磺胺胍具抗性的变异株Bc-66。在适宜的培养条件下,摇瓶发酵72h,可积累谷酰胺达31~33mg·mL-1,且遗传稳定性较好。对该菌株的发酵条件作了较系统的试验,摸索出较以前简化的培养基,有利于今后的进一步研究。
The selective breeding of an L glutamine producer and its fermentative conditions in shaking flask were studied.Mutant Bc 66 was derived from Brevibacterium flavum ATCC14067 by means of mutagenesis with diethyl sulphate(DES).The mutant showed a good character.Such as higher product of L glutamine,less by products and good resistance to sulfaguanidine(SG).It can accumulate 31~33 mg·mL -1 L glutamine after 72 hrs in shaking flask culture under optimum conditions.The fermentative medium and conditions were also discussed in this paper.
