非诺贝特对高脂诱导肥胖SD大鼠visfatin蛋白表达的影响 被引量:3

摘要 目的探讨过氧化物酶体增殖物激活受体-α(PPAR-α)激动剂非诺贝特对SD大鼠visfatin蛋白表达的影响。方法45只雄性SD大鼠适应性喂养1周后随机分为3组:普食组(NC组)、高脂组(HF组)、非诺贝特组(FF组)。NC组喂以普通饲料,其余两组给予高脂饮食。喂养12周后,FF组以非诺贝特30mg/(kg·d)灌胃治疗4周,空腹取血进行相关代谢指标测定,并检测腹部皮下脂肪、附睾脂肪和骨骼肌组织中visfatin蛋白表达。结果HF组大鼠体重(BW)、血清visfatin、空腹血糖(FBG)、空腹胰岛素(FINS)、游离脂肪酸(FFA)和甘油三脂(TG)明显高于NC组(P<0.05),非诺贝特能减少BW、下调血清visfatin表达(P<0.05),改善高脂血症和高胰岛素血症。Visfatin在附睾脂肪组织中表达显著高于皮下脂肪组织和肌肉组织,非诺贝特下调附睾脂肪组织visfatin表达。相关分析显示血清visfatin与FFA、TG、FBG呈正相关(P<0.05)。结论PPAR-a激动剂非诺贝特具有改善高脂饮食诱导的脂质异常、改善胰岛素抵抗以及下调visfatin作用。 Objective To examine the effect of peroxiseme protiferator-activated receptor-α(PPAR-α) agonist fenofibrate on visfatin expression in SD rats. Methods After 1 wk acclimation, 45 male SD rats were randomly assigned to 3 groups : normal control group ( NC group), high-fat diet group ( HF group) and fenofibrate group ( FF group). The animals of NC group were fed with normal chow and the others with high-fat diets. After 12 weeks, animals in FF group were treated with fenofibrate [ 30 mg/(kg ~ d) ] by intragastric administration for 4 wk. Then the blood of rats in fasting was obtained to measure metabolic parameters. Visfatin protein in abdominal subcutaneous adipose tissue,epididymis adipose tissue and muscle was quantitatively determined. Results Body weight (BW), serum visfatin, fasting blood glucose (FBG) ,fasting insulin (FINS),free fatty acid (FFA) and triglyceride (TG) of HF group were significantly higher than those of NC group (P 〈 0. 05) , and fenofibrate could down-regulate BW and serum visfatin ( P 〈 0. 05 ) and improve hyperlipemia and hyperinsulinemia. Visfatin expression was much higher in epididymis adipose tissue than in subcutaneous adipose tissue and muscle. Fenofibrate downregulated visfatin expression in epididymis adipose tissue (P 〈 0. 05). Additionally, we demonstrated that serum visfatin was positively correlated with FFA, TG and FBG (P 〈 0.05 ). Conclusion PPAR-αagonist fenofibrate may reverse high-fat diets induced lipid abnormalities, improve insulin resistance and down-regulate visfatin expression.
