摘要 尼克酰胺磷酸核糖转移酶(Nampt),又称为前B细胞克隆增强因子(PBEF)和内脏脂肪素(Visfatin),近年来由于其多方面的功能在尼克酰胺腺嘌呤二核苷酸(NAD)生物学、代谢、炎症等领域成为研究热点。作为NAD生物合成限速酶,Nampt调节NAD依赖蛋白如脱乙酰基酶的活性,并在胰岛β细胞分泌胰岛素中起重要作用。Nampt尚可作为具有免疫调节功能的炎症细胞因子,参与多种急慢性炎症性疾病的发病。本综述总结Nampt上述不同功能,并讨论其与2型糖尿病的关系。 Nicotinamide phosphoribosyltransferase( Nampt), which also called pre-B cell colony-enhancing factor(PBEF)and Visfatin, has recently drawn much attention in several fields, including nicotinamide adenine dinucleotide(NAD) biology, metabolism and inflammation because of its pleiotropic functions. As a rate-limiting NAD biosynthetic enzyme, Nampt regulates the activity of NAD-consuming proteins such as sirtuins, and plays an important role in regulating insulin secretion in pancreatic β cells. Nampt also serves as an immunomodulatory inflammatory cytokine and therefore, has a role in the pathogenesis of a variety of acute and chronic inflammatory diseases. This review summarized these various functional aspects of Nampt and discussed its relationship with type 2 diabetes.
