摘要
目的:探讨羟基脲(hydroxyurea,HU)对大鼠生精细胞凋亡相关基因mRNA及蛋白表达水平的影响。方法:雄性大鼠,腹腔注射400 mg.kg-1HU,给药后6,12和24 h处死,取睾丸组织,分离总RNA和蛋白。结果:半定量反转录聚合酶链式反应(RT-PCR)和蛋白印迹(Western blot)分析凋亡相关基因、蛋白产物,显示HU处理后,Bax水平没有出现明显变化,但Bcl-2表达水平在HU处理后6或12 h明显降低;相反,FasL和Fas的表达水平在给药后12 h开始增加。结论:HU导致的大鼠睾丸生殖细胞的凋亡涉及Bcl-2蛋白和Fas信号系统。
Objective:To determine the expression levels of apoptosis-related gene mRNA and protein in rat germ cells after hydroxyurea(HU) treatment.Methods: Adult male Wistar rats were injected(ip) with a single dose of HU(400mg·kg-1).The total RNA and protein were separated from the testis 6,12,or 24h after HU treatment.Results: RT-PCR and Western blotting were conducted for analysis of apoptosis-related gene and the protein products.The expression level of Bax did not change,but that of Bcl-2 significantly decreased at 6 or 12 h after HU treatment. In contrast, the expression levels of Fas and FasL significantly increased at 12 h. Conclusion: HU in- duces apoptosis of rat testicular germ cells, in which Bcl-2 protein and the Fas signaling system may be involved.
