摘要 目的利用突触蛋白Ⅰ(SynapsinⅠ,SYNⅠ)免疫组化染色,光镜下观察慢性砷中毒对成年大鼠海马CA3区突触蛋白的影响,探讨慢性砷中毒对成年大鼠脑的神经毒性。方法3~4月龄SD大鼠60只,体重(165±15)g,随机分为正常对照组(N,自由饮用蒸馏水)、低剂量组(L,As2O3按0.4mg/kg的比例用蒸馏水配制,自由饮用)和高剂量组(H,As2O3按10mg/kg的比例用蒸馏水配制,自由饮用)。每组20只,各组均用普通饲料喂养,连续喂养4个月后取大鼠海马组织,处死动物前进行血、尿生化指标的检测。常规石蜡切片,利用突触蛋白Ⅰ抗体进行SABC法免疫组化染色,观察大鼠海马CA3区突触蛋白的改变,并用图像分析软件测量NSE阳性反应产物的平均光密度。结果血、尿含砷量明显增高、突触蛋白数量减少,SynapsinⅠ免疫组化染色阳性反应产物平均光密度降低(P<0.05),并随浓度增加损害加重。结论慢性砷中毒可以导致大鼠海马CA3区突触蛋白数量减少,并随浓度增加损害加重。 Objective The use of synapsin I (Synapsin I, SYN I )immunohistochemical staining, light microscope observation of chronic arsenic poisoning in adult rat hippocampal area CA3 synapsin impact of chronic arsenic poisoning of the adult rat brain neurotoxicity. Methods 3--4-month- SD(Sprague- Dawley)rats 60 and weighing 150-180g, were randomly divided into four groups, normal control group (N, a free drink distilled water), low-dose group (L, As203 according to the proportion of 0.4mg/kg with distilled water preparation, and high-dose group (H, As2O3 according to the proportion of 10mg/kg prepared with distilled water, free drink). 20 in each group, the groups were fed with normal diet, fed for four months after the hippocampus of rats, the animals were killed before the blood and urine biochemical tests.Conventional paraffin sections, using antibodies to synapsin I SABC immunohistochernical staining method to observe the rat hippocampal area CA3 synapsin change, and measurement by image analysis software NSE-positive reaction products of the average optical density. Results Blood, urine arsenic increased significantly reduce the number of synaptic proteins, Synapsin I positive immunohistochemical staining reaction products to reduce the average optical density (P〈0.05), and with the increased concentration of heavier damage. Conclusion Chronic arsenic poisoning can lead to hippocampal area CA3 to reduce the number of synaptic proteins, and increased with the concentration of heavier damage.
