摘要
目的观察软脉灵口服液对实验性血管性痴呆(VaD)大鼠空间学习记忆能力和海马CA1区神经元凋亡的影响。方法选用永久性结扎双侧颈总动脉(2VO)制作VaD模型,用Morris水迷宫测定大鼠学习记忆能力,HE染色观察大鼠海马CA1区的形态学变化,TUNEL染色观察大鼠海马CA1区的神经元凋亡情况。结果与模型组比较,软脉灵高剂量组逃避潜伏期缩短(P<0.001)、1min穿越原平台所在位置次数增加(P<0.001);软脉灵高剂量组与低剂量组大鼠海马CA1区的凋亡细胞比模型组减少(P<0.01);与软脉灵低剂量组比较,高剂量组大鼠海马CA1区的凋亡细胞更少(P<0.01)。结论软脉灵口服液能改善拟VaD大鼠的空间学习记忆能力,减少拟VaD大鼠海马CA1区的凋亡细胞。这些作用呈一定的量效关系。
Objective To investigate the effect of Ruanmailing oral liquid on learning and memory and the neuronal apoptosis in the hippoeampal CA1 subregion in rats with vascular dementia(VaD). Methods To established a Vad rat model after chronic cerebral hypoperfusion caused by permanent bilateral common carotid arteries occlusion(2VO). Morris water maze experiment was adopted to test learning and momory in rats. Changes of morphous in the hippocampal CA1 subregion in rats were observed by hematoxylin and eosin(HE). The neuronal apaptosis was observed by in site end - labeling of fragmented DNA using the tereminal deoxynucleotidyl transferase reaetion(TUNEL). Results Escape latency was significantly shorten and the number of entries in the target area was significantly increased of rats in high dose group compared with those in model group(P〈0. 001). Compared with the model group, the quantity of the TUNEL positive cells was significantly decreased in the hippocampal CA1 subregion in Ruanmailing oral liquid high dose group and low dose group(P〈0.01). Compared with Ruanmailing oral liquid low dose group, the quantity of the TUNEL positive cells was decreased remarkabaly in the hippocampal CA1 subregion in Ruanmailing oral liquid high dose group(P〈0.01). Conclusion Ruanmailing oral liquid could improve the learning and memory in rats with vascular dementia, reduce the quantity of the TUNEL positive cells in the hippocampal CA1 subregion in rats with vascular dementia. It showed some dose - effect relationship.
