摘要
通过PCR扩增、克隆、序列测定获得29株鸭疫里默氏菌(RA)的16S rRNA和16S-23S rRNA间隔区的核苷酸序列.将16S rRNA和16S-23S rRNA间隔区序列连接后分析发现,1型菌株和大多数2型菌株分别处于不同的分支,而2型菌株中的RAf11、RAf3和RAf104不处在2型菌株的分支中,却与1型菌株非常接近.挑选RAf11菌株和2型分支中的RAf19菌株,分别制备灭活苗,免疫雏鸭后进行攻菌保护试验,RAf11菌株对1型菌株的攻击可产生较高的交叉保护率,达53.8%,而RAf19菌株对1型菌株攻击的保护率仅为23.1%.借助16S rRNA和16S-23S rRNA间隔区序列的分析,筛选到一株具交叉保护的RA菌株.
The sequences of 16S rDNA and 16S- 23S rDNA intergenic spacers of 29 strains of RiemereUa anatipestifer (RA) were determined. Phylogenetic analysis indicated that the serotype 1 strains and the majority of serotype 2 strains formed different branches, whereas serotype 2 strains RAfll, RAf3 and RAf104 were found to be closely related to serotype 1. Inactivated vaccines of RA serotype 2 were prepared using strain RAfl9 in serotype 2 branch and strain RAIl 1 in serotype 1 branch, respectively. After immunization, the ducks were challenged with serotype 1 and 2 strains. The vaccine prepared using strain RAIl 1 produced a higher degree cross-protection (53.8%) against challenge with serotype 1 strain. In contrast, the vaccine prepared using strain RAf19 produced cross-protection of 23.1% against challenge with serotype 1 strain. Using phylogenetic analysis of the sequences of 16S rDNA and 16S-23S rDNA intergenic spacers, a strain of RA with cross-protection was found.
