人参皂苷Rg3对HT-29细胞株MMP-1表达和迁移能力的影响 被引量:12

Effects of Ginsenoside Rg3 on Expression of Matrix Metalloproteinase-1 and Motile Ability of HT-29 Cell Line

摘要 目的:研究人参皂苷Rg3对结肠癌细胞株HT-29基质金属蛋白酶-1(MMP-1)表达和细胞迁移能力的影响。方法:体外培养HT-29细胞,分为人参皂苷Rg3高剂量组(100μg/mL)、中剂量组(50μg/mL)、低剂量组(25μg/mL)和空白对照组,培养HT-29细胞24、48、72h,RT-PCR法测MMP-1 mRNA表达。利用划痕实验观察Rg3对HT-29细胞迁移能力的影响。结果:24h高剂量组与对照组相比,有显著的统计学差异(P<0.01);48h低、中、高剂量组与对照组相比均有显著的统计学差异(P<0.05、P<0.01、P<0.01);72h低、中、高剂量组与对照组相比均有显著的统计学差异(P<0.01、P<0.01、P<0.01)。划痕实验中空白对照组过河时间为(47.33±2.49)h,低、中、高剂量组过河时间分别为(55.33±2.49)h(P<0.01)、(64.00±1.63)h(P<0.01)和(73.33±1.89)h(P<0.01)。结论:人参皂苷Rg3能抑制HT-29细胞MMP-1的表达和抑制HT-29细胞的迁移能力,并且随着药物浓度的增加及时间的延长,抑制作用增强。 Objective To investigate the effects of ginsenoside Rg3 on expression of mtrix metalloproteinas (MMP-1) and motile ability of colon cancer cell line HT-29. Methods HT-29 cell was divided into 4 groups:high dose group (ginsenoside Rg3 100 μg/mL),middle dose group (ginsenoside Rg3 50μg/mL),low dose group( ginsenoside Rg3 25μg/mL) and the control group.The expression of MMP-1 was tested by RT-PCR and motile ability was detected respectively by wound healing test at 24,48,72 h. Results In the 24 h groups:there were no significant differences among low-dosed,middle-dosed group and the control group.But there was significant difference between the high-dosed and the control group.In the 48 h groups:it showed statistical differences among low-/middle-/high dosed groups and the control group (P〈0.05,P〈0.01,P〈0.01).In the 72 h groups:the same result was found as in the 48 h groups.The wound healing time was 47.33±2.49 h,55.33±2.49 h,64.00±1.63 h,73.33±1.89 h respectively in control group,low-dosed,middle-dosed,high-dosed group,which showed statistical differences. Conclusion Ginsenoside Rg3 has obviously inhibited the expression and motile ability of HT-29 cell with the increase of time and concentration.
