远志皂苷对蛙坐骨神经干动作电位的影响

摘要 目的:观察远志皂苷对蛙坐骨神经兴奋性的影响。方法:用蛙制作坐骨神经标本,以1/3最大反应的刺激强度作为最适刺激强度,观察不同浓度远志皂苷(0.1 mg/mL、0.5 mg/mL、1 mg/mL和10 mg/mL)对坐骨神经动作电位幅值的影响;在获得最适合药物浓度的基础上,进一步观察高钾及低温环境下远志皂苷对蛙坐骨神经动作电位幅值的影响。结果:(1)各浓度的远志皂苷均能提高坐骨神经动作电位的幅值,分别为:(115.05±42.08)%,(136.60±44.75)%,(122.91±44.75)%及(120.23±65.41)%,以浓度为0.5 mg/mL时作用最显著(P<0.05);(2)高钾(1 mol/L)溶液与低温环境(10±2)℃分别使坐骨神经动作电位幅值降低为处理前的(16.31±10.81)%及(52.19±29.19)%,撤除高钾及低温条件则能恢复至基线水平.而0.5 mg/mL远志皂苷溶液却能使高钾及低温作用下的坐骨神经动作电位幅值得到提高,分别提高为基线的(118.47±42.42)%及(123.88±50.76)%,洗去远志皂苷后均能恢复至基线水平。结论:(1)远志皂苷能提高蛙坐骨神经的兴奋性,以0.5 mg/mL的效果最为明显;(2)高钾及低温环境对坐骨神经兴奋性具有抑制作用;(3)0.5mg/mL远志皂苷能改善高钾及低温对蛙坐骨神经兴奋性的抑制作用。 Objective: To study the effects of tenuigenin on excitability of sciatic nerve in frog. Method: The sciatic nerves from frogs were prepared and randomLy divided into four groups. The amplitudes of sciatic nerve of frog were been tested after treatment with different concentration of tenuigenin at 0.1 mg/mL,0.5 mg,/mL, 1 mg/mL and 10 mg/mL. After obtaining the suitable concentration of tenuigenin ,we observed the amplitudes of sciatic nerve of frog by using 1 moL/L KCI solution or low temperature treatment. Results: ( 1 ) The largest amplitude of sciatic nerve which used by the tenuigenin groups at the concentration of 0.1 mg/mL,0.5 mg/mL, 1 mg/mL and 10 mg/mL were ( 115.05 ±42.08)%, ( 136.60 ±44.75)% (P 〈0.05 ), ( 122.91±44.75 )% and ( 120.23± 65.41 )% ,respectively. (2) After 1 mol/L KCI solution or low temperature treatment, the amplitudes of sciatic nerve were been inhibited from 100% to ( 16.31± 10.81 ) % and ( 52.19 ± 29.19 ) %, respectively. And these then were improved to ( 118.47± 42.42 ) % and (123.88 ± 50.76)%, respectively, after using the tenuigenin at the concentration of 0.5 mg/mL. Conclusion: (1) the excitability of sciatic nerve could be improved by the tenuigenin; (2) the excitability of sciatic nerve were been inhibited by high KCI solution or low temperature; (3) the tenuigenin at the concentration of 0. 5mg/mL could improved the amplitudes of sciatic nerve which had been treatmented with high KC1 solution or low temperature.
