摘要
目的通过不同程度烧伤大鼠EPO(erythropoietin,促红细胞生成素)水平的变化,结合其烧伤程度以及血红蛋白(Hb)含量的降低程度,探讨EPO水平的变化规律及其与烧伤、贫血程度的相关性。方法将大鼠40只随机分为4组,每组10只,分别建立10%、20%及30%TBSAⅢ度烧伤大鼠模型及正常对照组。采用双抗体夹心ABC-ELISA法测定伤后12h、1d、2d、5d、7dEPO含量,并同期测定其血红蛋白含量,进行对照研究。结果伤后12h~7d,实验动物血清EPO浓度显著升高,12h即显著高于正常值,24h达到高峰,约为正常的4.5倍,此后逐渐下降,在烧伤后第5d仍显著高于正常,第7天后恢复至正常水平,且与烧伤程度呈正相关。烧伤大鼠血清EPO值与其血红蛋白含量呈负相关(r=-0.722P<0.05),即血红蛋白含量越低,血清EPO值越高。结论随着烫伤程度的增加,大鼠血清EPO水平逐渐升高,与烧伤程度呈正相关;随着血红蛋白的降低,大鼠血清EPO水平逐渐升高,与Hb呈负相关。这二种变化均由机体缺血缺氧造成。
Objective To investigate the rule of erythropoietin changed in serum and the relativity between erythropoietin and the degree of empyrosis and hemophthisis, erythropoietin and hemoglobin level was detected in burned rat with different degree. Methods Forty rats were divided into four groups randomly, one group was selected to be normal control, and the others were established as three-degree burned rat model with TBSA in 10% and 20% and 30% respectively. Double antibody sandwich ABC-enzyme-llnked immunosorbent assay (ELISA)was applied to detected the level of erythropoietin and hemoglobin at different time points such as 12 hours, 1 day,2 days,5 days and 7 days after rats had been burned. Results The burned rats' erythropoietin level was significant increased between 12 hours to 7 days. It was significant higher than normal level after 12 hours, and it reached the peak value which was 4. 5 folds compared with control group at 24 hours after the rats were burned. Then, the erythropoietin level declined gradually, it did not reach the normal level until 7 days. And the lower the hemoglobin content, the higher the erythropoietin level was. Conclusion There is positive relativity between the erythropoietin level and the burned degree, but negative relativity between the erythropoietin level and the hemoglobin content. Both changes are caused by body' s ischemia and hypoxia.
