结肠癌与正常结肠组织基因表达谱的差异性研究 被引量:3

摘要 目的:研究结肠癌与正常结肠黏膜组织的基因表达谱差异。方法:收集经病理证实的结肠腺癌组织和正常结肠黏膜组织各2例,抽提、纯化mRNA,制成荧光标记cDNA探针,与含有17101个cDNA的SBC人16K基因表达谱芯片杂交,计算机软件筛选差异表达基因。结果:与正常结肠组织比较,共筛选出1714条差异表达基因,其中表达上调有992条(57.88%),下调有722条(42.12%),差异表达最多的基因为与蛋白结合相关的基因,约占27.13%,其余还与代谢(18.49%)、细胞凋亡(16.74%)、细胞周期(9.92%)、信号转导(7.88%)及物质运输(4.96%)等多种作用有关。结论:基因表达谱芯片技术可以有效地筛选出结肠癌组织和正常结肠组织的差异表达基因,可应用于结肠肿瘤发病机制的研究。
