加味小柴胡颗粒对小鼠免疫性血小板减少性紫癜GR及其mRNA的影响 被引量:2

Influence of Modified Xiaochaihu Granules on expressions of GR and GR-mRNA in mice with immune thrombocytopenic purpura

摘要 目的观察糖皮质激素(GC)干预前后小鼠免疫性血小板减少性紫癜模型脾细胞糖皮质激素受体(GR)及其转录基因(GR-mRNA)的变化以及加味小柴胡颗粒的调控作用。方法建立小鼠免疫性血小板减少性紫癜模型,将之分为5组:正常组;模型组;加味小柴胡颗粒组;激素组;加味小柴胡颗粒+激素组。除正常组外,其余各组均造模,于造模后第7天开始分组给药,给药量为30mL/kg,每日给药1次,共14d。造模21d处死,取脾脏匀浆,放射配体结合分析法测定求出脾细胞GR位点数,实时荧光定量逆转录-聚合酶链反应(RT-PCR)法检测脾细胞GR-mRNA表达。结果小鼠免疫性血小板减少性紫癜模型脾细胞GR及mRNA下降,与正常组比较有显著差异(P<0.05);加味小柴胡颗粒组脾细胞GR及mRNA上升,与模型组比较有显著差异(P<0.05)。激素组指标下降更为明显,与模型组相比有显著差异(P<0.05);加味小柴胡颗粒+激素组则上升,与激素组比较有显著差异(P<0.05)。结论小鼠免疫性血小板减少性紫癜模型存在GR水平低下,并发生在mRNA水平,经激素干预后GR水平下降更显著,而加味小柴胡颗粒具有上调作用,且发生在mRNA水平。 Objective To observe the changes of glucocorticoid receptor (GR) and GR-mRNA in spleen cells before and after the intervene of glucocorticoid (GC) in mouse model of immune thrombocytopenic purpura and the regulation effect of Modified Xiaochaihu Granules (MXG). Methods All mice were divided into 5 groups, including the normal group, model group, MXG group, hormone group and MXG + hormone group. The mouse model of immune thrombocytopenic purpura was established in all groups except the normal group. After 7 days all groups were treated with the relevant drugs except the normal group (30 mL/kg, once a day for 14 days). After 21 days the mice were killed and the spleen was collected and nomogenized. The number of loci of spleen GR was deterimed with radioligand blinding assay, and the expression of GR-mRNA was detected with real-time fluorescence quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results The number of loci of GR and GR-mRNA expression decreased in the model group with a significant difference compared with those in the normal group ( P 〈 0.05 ), increased in the MXG group with a significant difference compared with those in the model group (P 〈 0.05 ), decreased more significantly in the hormone group with a significant difference compared with the model group (P 〈 0.05 ), increased in the MXG + hormone group with a significant difference compared with the hormone group (P 〈 0.05 ). Conclusion The number of loci of GR is lower, especially at the level of mRNA in mouse model of immune thrombocytopenic purpura. After the intervention by hormone the decrease of GR is more significant. Modified Xiaochaihu Granules can upregulate the number of loci of GR, which performs at the level of mRNA.
