摘要
目的:了解保健口服液的安全性。方法:设3个剂量组为12.5、25、50 ml/kg.BW,另设蒸馏水为对照组,以经口灌胃方式给予。试验期间每天观察动物形态,记录饲料洒漏量、体重、饲料和摄入量。灌胃30 d后,摘眼球采血作血常规及生化指标测定,取出肝、肾、脾、睾丸称重,并对肝、肾、脾、胃肠、睾丸、卵巢作病理学检查。结果:雌性各剂量组大鼠体重、雌性低、高剂量组体重增重低于对照组,差别有统计学意义(P<0.05);其余各组体重、体重增重、进食量、食物利用率与对照组比较,差别均无统计学意义(P>0.05)。雄性低剂量组血红蛋白、中剂量组丙转氨酶、谷草转氨酶,雌性中剂量组白细胞计数、高剂量组血糖与对照组比较差别有统计学意义(P<0.05),均在本实验室历史对照正常值范围内,一般认为无生物学意义。其余各组大鼠的血红蛋白、红细胞计数、白细胞计数和白细胞分类、血清谷丙转氨酶、谷草转氨酶、尿素、肌酐、总胆固醇、甘油三酯、血糖、总蛋白、白蛋白测定值与对照组相比,差别均无统计学意义(P>0.05)。对受检脏器作组织病理学检查,未见特异性病变。结论:保健口服液以12.5、25、50 ml/kg.BW剂量给大鼠灌胃,对雌性大鼠的体重、增重有一定影响,尚待研究,其余的未见不良的影响。
Objective:To know the safety of healthy oral liquid.Methods:All animals were divided into 4 groups.The control group of which was given water by gastric and other groups was given the oral liquid respectively with the dose of 12.5,25 and 50 ml/kg·BW.Observed the animals during the test every day and record the animal's weight,volume of intake and leakage of food about for 30 days.Then kill the animals and check blood for biochemical of blood and blood routine,take out the liver,kidney,spleen,stomach,testis to testis weight and have a pathological examination.Results:The body weight in all dose groups and weight gain in high group of female is lower than which in control group(P0.05),there is no significant difference between food intake,food utilization,body weight and weight gain in other dose groups with control group(P0.05);there are also no difference between biochemical of blood and blood routine in all dose groups with control group except the Hgb in high group,ALT and AST in middle dose group of male animals and WBC in middle dose group and Glu in high dose group of female animals,but they are all in range of the history compare value of our laboratory.Conclusion:Healthy oral liquid is not harmful to male rats when they were given this oral liquid by the dose of 12.5,25,50 ml/kg·BW but there is an effect on body weight of female rat.We will try to study it further.
